Multiomics profiling of cellular senescence in aging human skeletal muscle uncovers Maraviroc as a senotherapeutic approach for sarcopenia

Prof. Huating Wang, collaborating with Prof. Tim-yun Ong and Prof. Sun Hao (CUHK-Shenzhen), have recently published exciting research in Nature Communications. Their findings unlocked crucial insights into how cellular senescence drives skeletal muscle aging, paving the way for potential new treatments for sarcopenia.

As we age, skeletal muscle declines in mass, strength, and function. While cellular senescence, where cells stop dividing and secrete harmful factors, is implicated, its exact role in muscle aging has been unclear. In this study, using cutting-edge single-nucleus multi-omics technology, Prof. Wang’s team revealed increased cellular senescence in aging human skeletal muscle. They demonstrated the heterogeneity and dynamics of cellular senescence and uncovered how the senescent cells drive muscle aging through secreting SASP factors. Furthermore, their findings uncovered a novel senotheraupeutic target and a potential small molecule drug, Moroviroic for suppressing senescence and delaying muscle aging. Moreover, the study has also identified upstream transcription factor (TF) regulators of cellular senescence and dissected how one TF, JunB orchestrates SASP production. This work provides the first cellular senescence atlas in aging human muscle and establishes knowledge foundation for developing therapeutic strategies targeting sarcopenia. Dr. Yang Li and Dr. Chuhan Li are the co-first authors of this paper. The link to this publication is
